Tools for Genomic Applications Powered by CRISPR/CAS9 Based DNA Labeling

Overview

Executive Statement:

Drexel has a portfolio of tools and platforms for genomics applications based on the CRISPR/CAS9 system. The portfolio has applications in clinical diagnostics, cell and gene therapy, and genomics research among others. We are seeking licensing partners to translate the platforms and diagnostic tools for commercial use. We are also interested in partnership opportunities to develop applications not yet explored by our researchers since this portfolio has further innovation potential.

Description:

We have developed a novel CRISPR/Cas9-mediated labeling system that can be applied in whole genome mapping, target-specific mapping, DNA sequencing, and molecular diagnostic applications. Currently demonstrated applications include genome mapping and gene sequencing, on and off-target gene editing, telomere length detection and mapping, and rapid genomic characterization of tumor types.

The lab has developed a high accuracy, low cost, more efficient method for whole genome mapping and sequencing using a novel linked paired-end library generation method. It uses CRISPR/Cas9 nickase and sgRNAs enable non-random fragmentation of DNA (>300kb) into linked segments (~20 kb), preserving physical linkage information. Compatible with leading sequencing platforms, this method enhances targeted genomic studies.

We have also developed a novel platform for mapping and sequencing DNA on a micropatterned surface. The invention offers an innovative way to immobilize and optically map/sequence oligonucleotides on a micropatterned substrate for efficient large-scale genomic assembly.
